چکیده

Pre‐mRNA splicing requires the activities of small nuclear ribonucleoproteins and other essential splicing factors. Among these are members of the SR protein family and SR‐related proteins, which are integrally involved in regulating exon recognition, spliceosomal assembly, and spliceosomal re‐arrangements to promote intron excision. This chapter will focus on the discovery of SR proteins and SR‐related proteins, their structural organization, and their activities in mediating pre‐mRNA splicing. The importance of SR‐proteins in spliceosomal recruitment will be contrasted to other mechanisms resulting in alternative splicing. An additional aspect of the chapter will be a discussion on how reversible phosphorylation of the RS domain dictates SR protein activity and localization.

A novel SR-related protein is required for the second step of Pre-mRNA splicing.

The SR family proteins and SR-related polypeptides are important regulators of pre-mRNA splicing. A novel SR-related protein of an apparent molecular mass of 53 kDa was isolated in a gene trap screen that identifies proteins which localize to the nuclear speckles. This novel protein possesses an arginine- and serine-rich domain and was termed SRrp53 (for SR-related protein of 53 kDa). In suppor...

The SR protein family of splicing factors: master regulators of gene expression.

The SR protein family comprises a number of phylogenetically conserved and structurally related proteins with a characteristic domain rich in arginine and serine residues, known as the RS domain. They play significant roles in constitutive pre-mRNA splicing and are also important regulators of alternative splicing. In addition they participate in post-splicing activities, such as mRNA nuclear e...

The serine/arginine-rich protein family in rice plays important roles in constitutive and alternative splicing of pre-mRNA.

Ser/Arg-rich (SR) proteins play important roles in the constitutive and alternative splicing of pre-mRNA. We isolated 20 rice (Oryza sativa) genes encoding SR proteins, of which six contain plant-specific characteristics. To determine whether SR proteins modulate splicing efficiency and alternative splicing of pre-mRNA in rice, we used transient assays in rice protoplasts by cotransformation of...

Jumonji domain containing protein 6 (Jmjd6) modulates splicing and specifically interacts with arginine–serine-rich (RS) domains of SR- and SR-like proteins

The Fe(II) and 2-oxoglutarate dependent oxygenase Jmjd6 has been shown to hydroxylate lysine residues in the essential splice factor U2 auxiliary factor 65 kDa subunit (U2AF65) and to act as a modulator of alternative splicing. We describe further evidence for the role of Jmjd6 in the regulation of pre-mRNA processing including interactions of Jmjd6 with multiple arginine-serine-rich (RS)-domai...

The nuclear-retained noncoding RNA MALAT1 regulates alternative splicing by modulating SR splicing factor phosphorylation.

Alternative splicing (AS) of pre-mRNA is utilized by higher eukaryotes to achieve increased transcriptome and proteomic complexity. The serine/arginine (SR) splicing factors regulate tissue- or cell-type-specific AS in a concentration- and phosphorylation-dependent manner. However, the mechanisms that modulate the cellular levels of active SR proteins remain to be elucidated. In the present stu...
